Tango Therapeutics (TNGX) said Monday that initial results from its phase 1/2 trial of vopimetostat, an investigational PRMT5 inhibitor, in combination with Revolution Medicines' inhibitor daraxonrasib showed a six-month progression-free survival rate of 90% in patients with pancreatic cancer.As of May 28, 59 patients with previously treated MTAP-deleted and RAS-mutant pancreatic ductal adenocarcinoma or non-small cell lung cancer had been treated with a vopimetostat-based combination either daraxonrasib or zoldonrasib, the company said.In the vopimetostat plus daraxonrasib dose-escalation arm, patients received either 200 mg or 250 mg of vopimetostat once daily together with 100 mg of daraxonrasib once daily. The company reported a 92% objective response rate and a 90% six-month progression-free survival rate in patients with MTAP-deleted and RAS-mutant pancreatic ductal adenocarcinoma. Tango also reported a 100% objective response rate in patients with non-small cell lung cancer.In the vopimetostat plus zoldonrasib dose escalation arm, patients with PDAC received vopimetostat 200 mg or 250 mg QD plus zoldonrasib 600 mg or 1200 mg QD. Data showed a 52% of objective response rate and 74% six-month PFS rate.The vopimetostat plus daraxonrasib or combination combination were generally well tolerated across all dose levels, with no new safety signals observed, the company said.Tango said it plans in the second half of the year to finalize the design of a phase 3 randomized controlled trial of the combination approach in pancreatic cancer, release vopimetostat monotherapy data in lung cancer and initial TNG456 glioblastoma data, present second- and third-line pancreatic cancer combination data at a scientific conference, and begin a phase 1/2 study of vopimetostat in combination with ERAS-0015.Shares of the company were up more than 44% in premarket trading Monday.Price: $29.30, Change: $+9.08, Percent Change: +44.91%
